Comparison of Two Methods (Laser, Erythrocytes) to Induce an Experimental Increase of Intraocular Pressure in Rabbits

Purpose: To compare the efficacy of two experimental methods - argon laser trabeculoplasty and injection of preserved homologous erythrocytes - to induce an increased intraocular pressure in rabbits. Methods: Eleven female Chinchilla Bastard rabbits (Charles River, Kißlegg) (mean age 180±30 days, weight 1900-2200 g) were divided in 2 groups: in group 1 (n=5) the trabecular meshwork of the right eye was treated around 360 degrees with 655±141 laser burns (1.2 W Argon blue-green, 0.1 second duration, 50 micron spot size). In group 2, preserved homologous erythrocytes were prepared (Quigley 1980) and 1 ml injected in the anterior chamber of the right eyes. The observation time was for group 1 196±60 days and for group 2 158±44 days. Intraocular pressure was measured with the applanation tonometer ProTon (Tomey, Erlangen) in non-anesthetized rabbits (group 1: 79±9x, group 2: 67±7x). Results: No increase of intraocular pressure could be reached in group 1 (argon laser trabeculoplasty) in comparison to the untreated left eyes. However, in group 2 a statisticaly significant increase of intraocular pressure could be observed in the erythrocyte-treated eyes (p<0.038, student t-test). Conclusions: In comparison to the laser-treated eyes, erythrocyte-treated eyes in rabbits had stable and significant increased intraocular pressures over a long period. Therefore, this erythocyte-induced increase of intraocular pressure is a well established method to investigate glaucomatous processes.
